What the literature reports

CJC-1295 without DAC (also known as Modified GRF 1-29) is a 29-residue analog of growth-hormone-releasing hormone carrying four stabilizing substitutions: D-Ala at position 2, Gln at 8, Ala at 15, and Leu at 27, with a C-terminal argininamide. Removing the DAC (drug affinity complex) linker gives a much shorter-acting molecule than CJC-1295 with DAC.

Cellaire classifies CJC-1295 as clinical evidence for the DAC-conjugated parent, preclinical for the bare 1-29 peptide. Two randomized double-blind trials by Teichman et al. (JCEM 2006) in healthy adults reported 2- to 10-fold GH increases persisting ≥6 days and 1.5- to 3-fold IGF-I increases persisting 9–11 days with the DAC-conjugated form. The unconjugated 1-29 has no equivalent published dosing trial.

Molecular overview

CJC-1295 without DAC is the 29-residue peptide YADAIFTQSYRKVLAQLSARKLLQDILSR-NH2, a substituted analog of the endogenous GHRH(1-29) sequence.

The four substitutions (D-Ala2, Gln8, Ala15, Leu27) protect against DPP-IV and endopeptidase degradation, extending plasma stability from a few minutes to roughly 30 minutes without any albumin-tethering group.

CJC-1295 with DAC adds a maleimidopropionic acid group at the C-terminus that forms a covalent bond with plasma albumin, extending the half-life to days. The version sold as "CJC-1295 no DAC" omits that group entirely.

Neither variant is FDA-approved. Both appear on the FDA Category 2 bulk substances list.
